Cloning and Expression Analysis of PR4 in Brassica juncea var. tumida

Pathogenesis-related proteins (PRs) play an important role in the process of plant disease resistance. Among them PR4 is a member of PRs family and is an important in fl uencing factor acquiring systemic resistance in plant. A new PR4 gene was isolated with RACE technique from Brassica juncea var. tumida Tsen infected by Plasmodiophora brassicae. The gene was designated as BjPR4 (GenBank accession number: JX467703), containing 584 bp nucleotide and encoding 140 amino acids. BjPR4 showed high similarity with PR4 gene from other plants with a conserved Barwin domain, but without chitin binding domain, so it can be divided into PR4 I and PR4 II types. Phylogenetic analysis revealed the evolutionary conservation of these proteins among diverse species. qRT-PCR analysis of the BjPR4 expression pattern in roots of B. juncea var. tumida showed that BjPR4 transcribed at low-expression level in early stage inoculated with P. brassicae, and increased sharply in the fi nal course of the disease. After being treated with salicylic acid, BjPR4 expression in B. juncea var. tumida seedling roots increased shortly, peaking at 12 h, but then dropped down to low level. On the other hand, BjPR4 expression decreased after being treated with methyl jasmonic acid, ethylene and abscisic acid. The results suggested that BjPR4 transcript is regulated by different plant signal molecules. Expression characterization of BjPR4 in B. juncea var. tumida roots indicated that it might be involved in host resistance against pathogens. Fig 6, Tab 1, Ref 20
