The inhibitory effect of REGγ-knockout on cell infiltration

Abstract:
Peritoneal injections of Listeria Monocytogenes to wild-type and REGγ-knockout mice were performed for analyzing the roles of proteasome activator REGγ in immune cell infiltration. By survival assay, clone formation assay, flow cytometry and hematoxylin-eosin staining, we found that mice lacking REGγ shows significantly lower survival and Listeria Monocytogenes clearance. Furthermore, REGγ deficiency obviously inhibits the infiltration of neutrophil and monocyte to infection sites. Thus, the results suggest that REGγ deficiency suppresses immune cell recruitment to infection sites, resulting in impaired bacterial clearance. Rapid bacterial reproduction causes sepsis eventually, and leads to the reduced survival rate of REGγ-knockout mice.
